The Viceroy Francisco Toledo, during his visit to Arequipa was informed by the town council about his desire to found a monastery of nuns. This motivated him to grant the necessary licenses for the foundation of the "Monastery of Private Nuns of the Order of Santa Catalina de Siena". Years later, Doña María de Guzmán, widow of Diego Hernández de Mendoza, beautiful, rich and young woman who had no children, decides to seclude herself in the monastery under construction, giving her all her possessions. On September 10, 1579, the memory and capitulation of the foundation of the monastery signed between the Cabildo, Justicia y Regimiento of the City and Bishopric of Cusco was made, after signing the document, four lots, owned by the city, were delivered to the city. operation of the monastery and Doña María de Guzmán is named "first pobladorra and prioress of said Monastery".
On Sunday, October 2, 1580, a greater mass is celebrated in the city and Doña María, recognized as a founder, formally takes the habits.
The women who entered the monastery as nuns were criollas, mestizas and even daughters of curacas. Only in 1964 did the first Spanish nuns enter. The story also tells of the entrance of "poor nuns" and ladies of the city, who without embracing the religious life, entered the monastery to exercise their virtues.
In 1582, Arequipa endured a strong earthquake, suffering the construction of the monastery, serious damage. Despite this, the closure was preserved. Thereafter, a period of suffering began, in which the sisters themselves repaired their rooms. The chapel, due to lack of money, could not be renovated "for which the Blessed Sacrament was exposed in a straw hut".
